A practical reference on prothymosin alpha: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.
Reviewed 2025-08-25. Anything still debated is marked as such rather than presented as settled.
Immunological studies connect the peptide to multiple parts of the immune response. It has been reported to engage Toll-like receptor signaling, to promote dendritic cell maturation, and to influence the balance of T helper cell subsets. Changes in natural killer cell activity and in cytokine release appear in cell culture and animal models. These observations describe broad immunomodulatory behavior rather than a single defined receptor target, and the primary molecular interaction has not been settled.
Thymosin alpha-1 is a synthetic 28-amino-acid peptide whose sequence was first identified in extracts of bovine thymus tissue during the 1970s. The chain carries an acetyl group on its N-terminal serine. Its acidic residue content is high, which produces strong water solubility and an isoelectric point well below neutrality. Material supplied for laboratory and clinical use is manufactured by solid-phase peptide synthesis rather than purified from animal tissue. Different salt forms, such as the acetate, alter the counter-ion content without changing the peptide backbone.
Whether the free 28-residue peptide circulates in human tissue remains debated. The best-documented human source is prothymosin alpha, a larger acidic protein that carries the sequence at its N-terminus. Reports of measurable peptide levels in serum and lymphoid tissue exist, yet some of that signal may come from cross-reacting fragments or from the parent protein. Most reviews therefore treat prothymosin alpha as the established human molecule and describe independent circulation of the small peptide as an unresolved question.

Laboratory work indicates that the peptide acts on cells of both the innate and adaptive immune systems. Reported effects include signalling through Toll-like receptors on dendritic cells, enhanced T-cell maturation, and increased natural killer cell activity. These actions are described largely from cell-culture and animal experiments, and the precise receptor-level events remain incompletely defined. Studies in humans have generally measured immune markers rather than a single defined molecular target. The resulting picture remains partly descriptive.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Results across trials have been mixed, and several studies were small or conducted under differing protocols. Regulatory status varies by country, and the compound is not approved in every jurisdiction where it is studied. Evidence for any single indication should be read with attention to sample size and endpoint choice.
Thymosin alpha-1 is a 28-residue peptide first isolated from thymus tissue in the 1970s. It corresponds to the N-terminal portion of thymosin beta-4, from which it is cleaved in vivo. The peptide carries an acetyl group at its N-terminus, a modification that affects its charge and stability. Synthetic material produced by solid-phase peptide synthesis is chemically identical to the natural fragment and is the form used in research and clinical studies.
The peptide occurs naturally in thymic tissue and has been detected in serum and other biological fluids. Reported concentrations are low, and reliable measurement generally requires immunoassay or mass spectrometry with an enrichment step. It is released from a larger precursor, prothymosin alpha, by proteolytic cleavage, although the enzymes involved are not fully characterized. Whether circulating levels reflect thymic output specifically remains an open question.
Thymosin alpha 1 is a 28-amino-acid peptide first isolated from thymosin fraction 5, a bovine thymic extract. Its sequence begins with an acetylated serine residue and carries a high proportion of acidic residues, so the molecule has a net negative charge near neutral pH. Despite the shared name, it is unrelated in sequence to the thymosin beta family. Synthetic material prepared by solid-phase peptide synthesis is identical in sequence to the natural peptide.
The molecule consists of 28 amino acid residues with an acetyl group attached to the N-terminal serine. Its sequence is acidic overall, with several glutamic and aspartic acid residues distributed along the chain and no cysteine, so disulfide bridges do not form. The peptide carries a net negative charge at physiological pH. Because the N-terminus is blocked, the intact molecule resists degradation by many aminopeptidases, which contributes to its stability in biological fluids.
The peptide is generated in cells by cleavage of prothymosin alpha, a larger acidic protein encoded by the PTMA gene. Prothymosin alpha is expressed in many tissues, not only in the thymus, and its functions include nuclear roles in chromatin-related processes. The 28-residue fragment corresponds to the N-terminal portion of that precursor. How the cleavage occurs and how the fragment's concentration is regulated remain open questions; circulating amounts are small and difficult to measure reliably with routine assays.
The lyophilized peptide is a white to off-white powder that dissolves freely in water and in aqueous buffers near neutral pH. Because the molecule carries a net negative charge under physiological conditions, saline and phosphate solutions are the usual vehicles, while strongly acidic media are avoided. Stock solutions are commonly divided into small aliquots so that repeated freezing and thawing can be limited, since cycling may encourage aggregation. Solubility in organic solvents is poor and those solvents are seldom used as primary diluents.
Recommended storage for the dry powder is a freezer near minus twenty degrees Celsius, kept desiccated and away from light. Once dissolved, the peptide is less stable and is usually held at two to eight degrees Celsius for short intervals or frozen for longer storage. Stability studies focus on the acetylated terminus and the disulfide linkage because those features define the intact molecule. Common degradation routes include cysteine oxidation, deamidation of asparagine or glutamine side chains, and slow formation of higher-molecular-weight species.
While the Appalachian Highlands and Appalachian Uplands are generally continuous across the U.S./Canadian border, the St. Lawrence Valley area is handled differently in the physiographic classification schemas. The part of the St. Lawrence Valley in the United States is one of the second-level classifications, part of the Appalachian Highlands. In Canada, the area is part of the first-level classification, the St. Lawrence Lowlands. This includes the area around the city of Montreal, Anticosti Island, and the northwest coastline of Newfoundland. The dissected plateau area, while not actually made up of geological mountains, is popularly called "mountains", especially in eastern Kentucky and West Virginia, and while the ridges are not high, the terrain is extremely rugged. In Ohio and New York, some of the plateau has been glaciated, which has rounded off the sharp ridges and filled the valleys to some extent. The glaciated regions are usually referred to as hill country rather than mountains.
== Solar planets habitability == Water is necessary for life under all its forms presently known on Earth. Without water, microbial activity is not possible. Even if some micro-organisms can be preserved in the dry state (e.g., after freeze-drying), their growth is not possible without water. Micro-organisms also require sufficient space to develop. In highly compacted bentonite and deep clay formations, microbial activity is limited by the lack of space and the transport of nutrients towards bacteria and the elimination of toxins produced by their metabolism is controlled by diffusion in the pore water. So, "space and water restrictions" are two limiting factors of the microbial activity in deep sediments. Early biotic diagenesis of sediments just below the ocean floor driven by microbial activity (e.g., of sulfate reducing bacteria) end up when the degree of compaction becomes too important to allow microbial life development. At the surface of planets and in their atmosphere, space restrictions do not apply, therefore, the ultimate limiting factor is water availability and thus the water activity. Most extremophile micro-organisms require sufficient water to be active. The threshold of water activity for their development is around 0.6. The same rule should also apply for other planets than Earth.
== History == Tetraoxygen was first predicted in 1924 by Gilbert N. Lewis, who proposed it as an explanation for the failure of liquid oxygen to obey Curie's law. Though not entirely inaccurate, computer simulations indicate that although there are no stable O4 molecules in liquid oxygen, O2 molecules do tend to associate in pairs with antiparallel spins, forming transient O4 units. In 1999, researchers thought that solid oxygen in its ε-phase, also known as red oxygen, (at pressures above 10 GPa) was O4. However, in 2006, it was shown by X-ray crystallography that this stable phase is in fact octaoxygen (O8). Nevertheless, positively charged tetraoxygen has been detected as a short-lived chemical species in mass spectrometry experiments.
== Diagnostic use == S100B is secreted by astrocytes or can spill from injured cells and enter the extracellular space or bloodstream. Serum levels of S100B increase in patients during the acute phase of brain damage. Over the last decade, S100B has emerged as a candidate peripheral biomarker of blood–brain barrier (BBB) permeability and CNS injury. Elevated S100B levels accurately reflect the presence of neuropathological conditions including traumatic head injury or neurodegenerative diseases. Normal S100B levels reliably exclude major CNS pathology. Its potential clinical use in the therapeutic decision making process is substantiated by a vast body of literature validating variations in serum 100B levels with standard modalities for prognosticating the extent of CNS damage: alterations in neuroimaging, cerebrospinal pressure, and other brain molecular markers (neuron specific enolase and glial fibrillary acidic protein). However, more importantly, S100B levels have been reported to rise prior to any detectable changes in intracerebral pressure, neuroimaging, and neurological examination findings. Thus, the major advantage of using S100B is that elevations in serum or CSF levels provide a sensitive measure for determining CNS injury at the molecular level before gross changes develop, enabling timely delivery of crucial medical intervention before irreversible damage occurs. S100B serum levels are elevated before seizures suggesting that BBB leakage may be an early event in seizure development.
Mendelevium was the ninth transuranic element to be synthesized. It was first synthesized by Albert Ghiorso, Glenn T. Seaborg, Gregory Robert Choppin, Bernard G. Harvey, and team leader Stanley G. Thompson in early 1955 at the University of California, Berkeley. The team produced 256Md (half-life 77.7 minutes) when they bombarded an 253Es target consisting of only a billion (109) einsteinium atoms with alpha particles (helium nuclei) in the Berkeley Radiation Laboratory's 60-inch cyclotron, thus increasing the target's atomic number by two. 256Md thus became the first isotope of any element to be synthesized one atom at a time. In total, seventeen mendelevium atoms were detected. This discovery was part of a program, begun in 1952, that irradiated plutonium with neutrons to transmute it into heavier actinides. This method was necessary because of a lack of known beta decaying isotopes of fermium that might allow production by neutron capture; it is now known that such production is impossible at any possible reactor flux due to the very short half-life to spontaneous fission of 258Fm and subsequent isotopes, which still do not beta decay - the fermium gap that, as far as we know, sets a hard limit to the success of neutron capture processes.

In the US, newborn screening (NBS) is mandated in all states, though in some states parents may decline it for religious or philosophical reasons. Few parents opt for the program due to health concerns, and a lack of awareness of the ability to opt out. After the initial testing is complete, the residual dried blood spots (DBS) on newborn screening cards may be used for secondary purposes, including being shared with law enforcement and sold for research. The decreasing costs of whole genome sequencing have also raised concerns that blood spots may be sequenced in the future, limiting any de-identification procedures. While CLIA does specify minimum retention requirements, it does not specify a federal maximum retention period. Retention periods for NBS cards vary by state; for example, New Jersey stores them for 23 years, and Texas may keep them indefinitely. The absence of parental awareness and consent for these activities, and a lack of transparency and federal regulations, has led to significant public concern and apprehension.
== Applications of commercially synthesized peptides == Biologically active peptides have been integrated into a growing number of active pharmaceutical ingredients (API’s) as well as standalone products such as vasopressin, gonadorelin, leuprolide, and goserelin. Completion of the Human Genome Project resulted in the identification of approximately 30,000 proteins encoded in the human genome and provided many more new target molecules for biomedical researchers to explore. To investigate the possibility of increasing the native potency of a given peptide or protein using a rational design approach, small and large amounts of peptides are needed, Some in the milligram scale. Once a desired activity or potency is identified, larger scale synthesis is need. For this, gram to multi-gram scale may be needed in order to initiate small animal studies. Often, after successful validation, an even larger scale of synthesis may be desired. These can range in scale from hundreds of grams to multi-kilo amounts.
While acutely effective however, tachyphylaxis has been found to occur with continuous administration of atomoxetine. The reasons for this tolerance are unclear. Though development for Alzheimer's disease was discontinued, perhaps due to negative trial findings, there has been continued interest in atomoxetine in the potential treatment of this disease as of 2025. It has also been studied to treat mild cognitive impairment or prodromal Alzheimer's disease. Atomoxetine is being studied for treatment of sleep apnea in combination with various other drugs including oxybutynin or aroxybutynin, trazodone, antimineralocorticoids like spironolactone, an orexin receptor antagonist, pimavanserin, acetazolamide, dronabinol (Δ9-tetrahydrocannabinol; THC), fesoterodine, and zolpidem. Atomoxetine has been studied for reducing appetite and promoting weight loss in people with obesity, with mixed results.

In July 2015, Allergan agreed to sell its generic drug business (Actavis Generics) to Teva for $40.5 billion ($33.75 billion in cash and $6.75 billion worth of shares). As a result, Teva dropped its pursuit of Mylan. In order for the deal to gain regulatory approval, Teva sold off a number of assets, including a portfolio of five generic drugs to Sagent Pharmaceuticals for $40 million, as well as a further eight medicines to Dr. Reddy's in a $350 million deal. Teva also sold a further 15 marketed generics, as well as three others that were close to market, for $586 million to Impax Laboratories. In July, Teva sold off a further 42 products to Australian generics company, Mayne Pharma, for $652 million; the deal moved Mayne up 50 spots, into the top-25 companies of US generic companies. As part of the deal Teva will seek to raise $20 to $25 billion through a bond sale. After completing the $39 billion acquisition of Actavis Generics, Teva announced another, smaller, deal with Allergan, agreeing to acquire its generic distribution business Anda for $500 million.

The TOPS System (Total Posterior Spine System) is a spinal implant designed to stabilize the lower spine and maintain range of motion following lumbar decompression surgery for spinal stenosis and related conditions. Unlike spinal fusion, which eliminates motion at the treated level, TOPS aims to preserve mobility while providing stability. The device is manufactured by Premia Spine and has received approval for clinical use in the United States by the Food and Drug Administration (FDA) and in the European Union under the CE (Conformité Européenne) marking.
===== Endopeptidases ===== Endopeptidases are enzymes that add water to an internal peptide bond in a peptide chain and break that bond. Three common endopeptidases that come from the pancreas are pepsin, trypsin, and chymotrypsin. Chymotrypsin performs a hydrolysis reaction that cleaves after aromatic residues. The main amino acids involved are serine, histidine, and aspartic acid. They all play a role in cleaving the peptide bond. These three amino acids are known as the catalytic triad which means that these three must all be present in order to properly function. Trypsin cleaves after long positively charged residues and has a negatively charged binding pocket at the active site. Both are produced as zymogens, meaning they are initially found in their inactive state and after cleavage though a hydrolysis reaction, they becomes activated. Non-covalent interactions such as hydrogen bonding between the peptide backbone and the catalytic triad help increase reaction rates, allowing these peptidases to cleave many peptides efficiently.
Xaa-Pro dipeptidase, also known as prolidase, is an enzyme that in humans is encoded by the PEPD gene. Prolidase is an enzyme in humans that plays a crucial role in protein metabolism and collagen recycling through the catalysis of the rate-limiting step in these chemical reactions. This enzyme is coded by the gene PEPD (peptidase D), located on chromosome 19. Serum prolidase activity is also currently being explored as a biomarker for diseases.

It is usually described as an immunomodulatory peptide rather than a classic circulating hormone. No endocrine gland is known to release it as a primary secretory product, and its measured presence in blood is not firmly established.
Prothymosin alpha is a much larger acidic protein, roughly 111 to 113 residues long, and the thymosin alpha-1 sequence matches its N-terminal region. The small peptide is therefore best understood as a fragment of that parent protein rather than a separate gene product.
Trials and clinical reports have examined chronic hepatitis B and C, use as a vaccine adjuvant, and supportive treatment in some immunodeficiency and oncology settings. Results vary by indication, and regulatory approval differs between countries.
