Thymosin alpha 1 is a short peptide first isolated from bovine thymus tissue in the early 1970s during fractionation work aimed at identifying factors that influence T cell development. It belongs to a family of acidic thymic peptides, and the original preparations contained several components that were later separated by chromatography. The compound is now produced synthetically rather than extracted from tissue, which removes batch variability tied to animal sourcing. Researchers describe it as an immunomodulatory peptide because laboratory studies show effects on several cell types of the innate and adaptive immune systems.
The molecule consists of 28 amino acid residues with an acetyl group attached to the N-terminal serine. Its sequence is acidic overall, with several glutamic and aspartic acid residues distributed along the chain and no cysteine, so disulfide bridges do not form. The peptide carries a net negative charge at physiological pH. Because the N-terminus is blocked, the intact molecule resists degradation by many aminopeptidases, which contributes to its stability in biological fluids.

Most published studies on thymosin alpha-1 report changes in immune measurements rather than clinical outcomes, and findings differ across designs and populations. Whether the peptide signals through one defined receptor or through several less specific interactions remains an open question. Its reported circulation half-life of a few hours complicates comparison of dosing schedules across trials. Mechanistic claims are frequently drawn from isolated cell cultures, and how far those results extend to whole organisms is unresolved.
Thymosin alpha-1 is a synthetic peptide of 28 amino acids whose sequence matches the amino-terminal region of prothymosin alpha. The chain is acetylated at its first residue and contains one disulfide bridge between two cysteine residues, which folds the molecule into a compact loop. Its molecular formula, C129H215N33O55, corresponds to a monoisotopic mass of roughly 3,106 daltons. Material used in laboratories is made by solid-phase synthesis rather than isolated from animal tissue.
Early work on thymic extracts in the 1960s described a heat-stable acidic fraction containing many polypeptides. Separation of that mixture yielded individual components, and thymosin alpha-1 was named as one of them on the basis of assays for T-cell activity. The first preparations came from calf thymus, while subsequent research and clinical material has been chemically synthesized. Nomenclature in older papers is inconsistent, and the same peptide sometimes appears under different designations, which complicates literature searches.
The lyophilized peptide is generally stable for extended periods when kept cold and dry. Once dissolved, aqueous solutions are less stable; hydrolysis, oxidation, and aggregation can degrade the material. Storage at -20 °C or lower slows these processes. Repeated freeze-thaw cycles are best avoided because they can promote aggregation. The exact shelf life depends on formulation, pH, and concentration, so stability studies are typically performed for each specific product.
Quality control for thymosin alpha-1 focuses on identity, purity, and potency. Identity is confirmed by mass spectrometry and amino acid analysis, while purity is assessed by chromatography with limits on related substances and residual solvents. Potency assays may use cell-based immune readouts, but these are not standardized across laboratories. Regulatory status differs by jurisdiction; no product is approved in the United States for clinical use, whereas some other countries register injectable forms for specific indications.
== Prognosis == Androgenic alopecia is typically experienced as a "moderately stressful condition that diminishes body image satisfaction". However, although most men regard baldness as an unwanted and distressing experience, they usually can cope and retain integrity of personality. Although baldness is not as common in women as in men, the psychological effects of hair loss tend to be much greater. Typically, the frontal hairline is preserved, but the density of hair is decreased in all areas of the scalp. Previously, it was believed to be caused by testosterone, just as in male baldness, but most women who lose hair have normal testosterone levels.
The four substrates of this enzyme are 1-hydroxy-2-naphthoic acid, reduced nicotinamide adenine dinucleotide (NADH), oxygen, and a proton. Its products are 1,2-naphthalenediol, oxidised NAD+, water, and carbon dioxide. It is involved in chrysene degradation in some bacteria.
Sometimes called "non-genetic EDS", hypermobility spectrum disorder can have the same signs as hEDS, but be caused not by a heritable genetic mutation but by problems in fetal development, such as pre-natal exposure to toxins like agricultural chemicals, drugs, or alcohol. Fetal Alcohol Spectrum Disorders affect at least 1 in 20 people in the U.S., and joint hypermobility with other symptoms is common.
In sensory neurons, an external signal such as pressure, temperature, light, or sound is coupled with the opening and closing of ion channels, which in turn alter the ionic permeabilities of the membrane and its voltage. These voltage changes can again be excitatory (depolarizing) or inhibitory (hyperpolarizing) and, in some sensory neurons, their combined effects can depolarize the axon hillock enough to provoke action potentials. Some examples in humans include the olfactory receptor neuron and Meissner's corpuscle, which are critical for the sense of smell and touch, respectively. However, not all sensory neurons convert their external signals into action potentials; some do not even have an axon. Instead, they may convert the signal into the release of a neurotransmitter, or into continuous graded potentials, either of which may stimulate subsequent neuron(s) into firing an action potential. For illustration, in the human ear, hair cells convert the incoming sound into the opening and closing of mechanically gated ion channels, which may cause neurotransmitter molecules to be released. In similar manner, in the human retina, the initial photoreceptor cells and the next layer of cells (comprising bipolar cells and horizontal cells) do not produce action potentials; only some amacrine cells and the third layer, the ganglion cells, produce action potentials, which then travel up the optic nerve.

=== Papillitis/hypertrophy === Papillitis refers to inflammation of the papillae, and sometimes the term hypertrophy is used interchangeably. In foliate papillitis the foliate papillae appear swollen. This may occur due to mechanical irritation, or as a reaction to an upper respiratory tract infection. Other sources state that foliate papilitis refers to inflammation of the lingual tonsil, which is lymphoid tissue.
=== Europe === According to regulations applicable in the European Economic Area two medicinal products are bioequivalent if they are pharmaceutically equivalent or pharmaceutical alternatives and if their bioavailabilities after administration in the same molar dose are similar to such a degree that their effects, with respect to both efficacy and safety, will be essentially the same. This is considered demonstrated if the 90% confidence intervals (90% CI) of the ratios for AUC0–t and Cmax between the two preparations lie in the range 80–125%.
== Pathophysiology == Ochronosis occurs because of deposition of phenols (such as homogentisic acid and hydroquinone) as plaques in the matrix of cartilage. The pigments can also be incorporated into collagen and elastin fibers. In the skin, the pigment alters the structure of the fibers, causing enlargement and curling. The embedded pigments also form crosslinks with pigment depositions in adjacent fibers, stabilizing and reducing the elastic recoil of the fibers. This results in hardening of elastic structures, increasing their rigidity and brittleness. Once ruptured, the exposed pigments cause a foreign body reaction and inflammation. This pigment deposition also invokes deposition of hydroxyapatite, the mineral responsible for bone calcification, further hardening the connective tissue. The pigment can also be excreted by glandular cells in apocrine and ceruminous sweat glands, as well as breast and prostate tissue. This results in darkly pigmented sweat and breast milk. Excretion of the pigment is only found in endogenous ochronosis and should not occur from topical phenols.
In 2022, he rejoined his former departments at Harvard and the Dana–Farber Cancer Institute. Cantley is married to Vicki Sato, herself a prominent figure in the pharmaceutical industry and a professor at Harvard University in both the Business and Medical Schools.
==== Imaging ==== Ultrasound of the thyroid may be used to reveal whether structures are solid or filled with fluid, helping to differentiate between nodules and goitres and cysts. It may also help differentiate between malignant and benign lesions. When further imaging is required, a radiolabelled iodine-123 or technetium-99 uptake scan may take place. This can determine the size and shape of lesions, reveal whether nodules or goitres are metabolically active, and reveal and monitor sites of thyroid disease or cancer deposits outside the thyroid. A fine needle aspiration of a sample of thyroid tissue may be taken in order to evaluate a lesion seen on ultrasound which is then sent for histopathology and cytology. Computed tomography of the thyroid plays an important role in the evaluation of thyroid cancer. CT scans often incidentally find thyroid abnormalities, and thereby practically becomes the first investigation modality.

=== Identification of de novo emerging sequences === There are two major approaches to the systematic identification of novel genes: genomic phylostratigraphy and synteny-based methods. Both approaches are widely used, individually or in a complementary fashion. To standardise terminology for translated non-canonical ORFs (often implicated in de novo gene studies), a community proposal introduced the term "translons" to denote all translated regions detected by approaches such as ribosome profiling.
=== David Healy === David Maurice Healy is played by Johnny Galecki. David is Mark Healy's younger brother. He first appeared in "The Bowling Show" (episode 4.14). Whereas Mark was initially a rebellious delinquent, David is friendly and respectful to the Conners. He is shy, polite, thoughtful, sensitive, soft-spoken, artistic, and intelligent. Much of his passive behavior results from his difficult home life and his mother's abusive behavior. He is romantically involved with Darlene, becoming the submissive partner. They also collaborate artistically, with David illustrating graphic novels that Darlene writes. His sweet manner endears him to the Conners, who eventually consider him as part of the family and jokingly refer to him as being more welcome than Darlene. He moves in with the Conners after Roseanne, herself a victim of child abuse, witnesses how abusive David's mother is. David and Darlene break up three times throughout the series, each for a successively longer period of time, but always reunite. After Darlene becomes pregnant, she and David married. In the series finale, Roseanne reveals in her writing that David had actually been dating Becky in "real life", and that Roseanne simply wrote his relationship as being with Darlene because she felt it made more sense. The series finale is retconned for season 10, where it is revealed that David and Darlene have been separated for some years and David has been away, traveling and doing volunteer work abroad, leaving Darlene to raise their two children, Harris and Mark, as a single mother.
==== Substrate or product ==== Substrate or product inhibition is where either an enzymes substrate or product also act as an inhibitor. This inhibition may follow the competitive, uncompetitive or mixed patterns. In substrate inhibition there is a progressive decrease in activity at high substrate concentrations, potentially from an enzyme having two competing substrate-binding sites. At low substrate, the high-affinity site is occupied and normal kinetics are followed. However, at higher concentrations, the second inhibitory site becomes occupied, inhibiting the enzyme. Product inhibition (either the enzyme's own product, or a product to an enzyme downstream in its metabolic pathway) is often a regulatory feature in metabolism and can be a form of negative feedback.
Smoking leads to an increased risk of bone fractures, especially hip fractures. It also leads to slower wound healing after surgery, and an increased rate of postoperative healing complication. Tobacco smokers are 30-40% more likely to develop type 2 diabetes than non-smokers, and the risk increases with the number of cigarettes smoked. Furthermore, diabetic smokers have worse outcomes than diabetic non-smokers.

Thymosin fraction 5 is a mixture of many peptides obtained from thymus tissue, while thymosin alpha 1 is a single defined 28-residue molecule. The two names appear together in older literature because the purified peptide was first obtained from that mixture.
The precursor, prothymosin alpha, is encoded by the PTMA gene in humans. The 28-residue peptide corresponds to the N-terminal segment released from that larger protein.
It is produced as a fragment of prothymosin alpha and is present in many tissues. Circulating concentrations are low, which makes routine measurement difficult.
它促进未成熟T细胞分化并增强成熟T细胞的增殖与细胞因子分泌。这些作用有助于放大抗原特异性免疫应答。
